A. Handling a Microscope

1. Inspect the parts if complete and functional.
2. Clean with tissue or cotton cloth the metal parts and lens paper for the lenses.
3. Put down gently the microscope about 2 inches away from the edge of the table.
4.Carry the microscope in an upright position to your working station.
5. Grab the arm with one hand and the base on the other hand.

B. Preparing a Wet Mount

1. Add a drop of water over the specimen (water act as a mounting medium).
2. Clean the microscope slide and cover slip.
3. Slowly drop the coverslip supported by a pin until it rests on the water and printed letter.
4. Position the cover slip 45° with one side touching one edge of the water.
5. Cut a small letter "e" from a newsprint and place it on the slide using a tweezer or forceps.

C. Setting Up a Microscope

1. Secure the slide using a stage clip and adjust the stage to center the specimen on the aperture.
2. Rotate the revolving nosepiece and set the LPO (a click sound is heard once the objective is in position).
3. Turn on the illuminator or adjust the mirror for proper lightning (never use direct sunlight).
4. Raise the stage as you look at the side by turning the coarse adjustment knob.
5. Place the slide on the stage.

D. Focusing and Observing Specimen Under a Microscope

1. Turn the coarse adjustment knob upward to get a view of the specimen.
2. Look through the eyepiece with both eyes open.
3. Sharpen the focus by turning the fine adjustment knob.
4. Shift the objective by rotating the nosepiece to HPO for detailed view.
5. Move the slide up and down, side to side to get better image of the specimen.​
